I used both Matchmaker and Hormone (Separately on the same save outside Riverwood a day after breaking out of Helgen) to get my character to masturbate and it doesn't animate. For matchmaking, nothing happens the first time while the following ones give a fizzle. For hormone, the sound goes off and a couple seconds later semen is visible on her vaginal area (despite having female cum turned off).

Do you have Sexlab aroused installed as well?  I think comfort needs that.

EDIT: IT is also possible that lovers hook is blocking animations you have not yet learned.  Try it without. (Or disable in MCM)
